THE ROLE OF VITAMIN B,j IN THE SYNTHESIS OF TRYP- TOPHANE FROM INDOLE AND ANTHRANILIC ACID BY LACTOBACILLUS ARABINOSUS”

چکیده
Lactobacillus arabinosus has the ability to synthesize several amino acids when adequate quantities of vitamin Bg are included in the medium (1, 2) ; however, the requirement of this organism for tryptophane is apparently not affected by the addition of vitamin 136 (2). Earlier studies showed that indole and anthranilic acid have appreciable tryptophane activity for L. arabinosus (3-7) but not for Streptococcus faecalis R (3,7). Serine enhanced the activity of indole for L. arabinosus when tested with a purified amino acid medium, but no increase in the tryptophane activity was noted when serine was added to a casein hydrolysate medium (7). Tatum and associates (8-10) have observed that serine is necessary for the synthesis of tryptophane from indole by certain Neurospora mutants, while Umbreit et al. (11) have shown that pyridoxal phosphate functions as a coenzyme for Neurospora in the synthesis of tryptophane from indole. In the present study the influence of pyridoxine, pyridoxal, and pyridoxamine and other constituents of the medium on the biosynthesis of tryptophane from indole and anthranilic acid by Lactobacillus arabinosus was investigated.

HE tryptophan-I ( t r yp I ) locus is located in linkage group I11 in Neurospora crassa. TATUM and BONNER (1944) and BEADLE and TATUM (1945) reported tryp-I mutant 10575, which could utilise either indole or tryptophan. AHMAD and CATCHESIDE (1960) brought forward evidence that a new group of five anthranilic acid mutants were allelic to 10575. The present paper describes complementation studies...
متن کاملAn isotopic study of the conversion of anthranilic acid to indole.
Anthranilic acid and indole have been implicated as intermediates in the biosynthesis of tryptophan in several microorganisms (l-5). A mechanism of tryptophan synthesis from indole has been established, namely the pyridoxal phosphate-dependent coupling of indole and L-serine (6-9). Previous studies of the conversion of anthranilic acid to the indole ring of tryptophan have shown that the amino ...

Rydon (1) has found that 4and 5-methylanthranilic acids inhibit the growth of Eberthella typhi and that this inhibition is reversed by anthranilic acid, indole, and tryptophan. He concluded that the two methyl compounds interfere with the synthesis of tryptophan at the stage of anthranilic acid. Interruption in the conversion of anthranilic acid to indole in Escherichia coli, caused by chloromy...

The accumulation of imidazoleglycerol phosphate during growth of Neurospora crassa in the presence of 3-amino-1,2,4-triazole was found to cause derepression of tryptophan synthetase and to inhibit the induction of kynureninase. Accumulation of indoleglycerol phosphate in response to growth in the presence of indole acrylic acid or anthranilic acid was also accompanied by derepressed synthesis o...
